Selank, developed by the Institute of Molecular Genetics of the Russian Academy of Sciences, is a nootropic and anxiolytic peptide-based medication. Selank is a heptapeptide with the sequence Thr-Lys-Pro-Arg-Pro-Gly-Pro (TKPRPGP), serving as a synthetic analogue of human tuftsin.

Pharmacology: Selank mimics many effects of the immunomodulatory peptide tuftsin as a synthetic analogue. It regulates the expression of Interleukin-6 (IL-6) and affects the balance of T helper cell cytokines. Additionally, it influences the concentration of monoamine neurotransmitters and induces serotonin metabolism. Selank rapidly increases the expression of brain-derived neurotrophic factor (BDNF) in the hippocampus of rats.

Selank, along with the related peptide drug Semax, inhibits enzymes involved in the degradation of enkephalins and other endogenous regulatory peptides, possibly contributing to their effects. It also affects the activity of carboxypeptidase H and phenylmethylsulfonylfluoride-inhibited carboxypeptidase in rat nervous system tissue.

Selank exhibits antidepressant-like effects in animal models of depression and anhedonia.

Clinical Trials: In clinical trials, Selank has demonstrated a sustained nootropic and anxiolytic effect, beneficial for treating generalized anxiety disorder (GAD). It offers advantages over traditional anxiety treatments like benzodiazepines, as it lacks sedating or negative cognitive side effects, as well as associated addiction or withdrawal issues.

Selank is closely associated with Semax, another nootropic drug developed by the Institute of Molecular Genetics in Russia. Semax is currently available in Russian and Ukrainian pharmacies.
